Pressure overload-induced heart failure was induced in non-transgenic B6 mice and apoE-deficient mice by abdominal aortic constriction (AAC). All mice had free access to food and water. To induce atherosclerosis-dependent heart failure apoE-deficient mice were fed a rodent chow containing 7 % fat and 0.15 % cholesterol (Ain 93-based diet). All other mice were fed a standard rodent chow containing 4.5 % fat. At an age of 18 months, hearts were isolated from apoE-deficient mice with symptoms of heart failure (ejection fraction ≤33% as determined by echocardiography) and age-matched B6 controls. Failing hearts from aortic constricted mice were isolated from (i) 6 month-old apoE-deficient mice with 2 months of AAC, and from (ii) 10 month-old B6 mice with 6 months of AAC (ejection fraction ≤33 %). As controls, heart tissue from age-matched, sham-operated apoE-deficient and non-transgenic B6 mice were used. Hearts from 5 month-old B6 mice with 1 month of AAC were also included in the study displaying cardiac hypertrophy without failure (ejection fraction ≥50 %).
