To induce atherosclerosis-dependent heart failure apoE-deficient mice were fed a rodent chow containing 7 % fat and 0.15 % cholesterol (Ain 93-based diet). All other mice were fed a standard rodent chow containing 4.5 % fat. At an age of 18 months, hearts were isolated from apoE-deficient mice with symptoms of heart failure (ejection fraction ≤33% as determined by echocardiography) and age-matched B6 controls. Failing hearts from aortic constricted mice were isolated from (i) 6 month-old apoE-deficient mice with 2 months of AAC, and from (ii) 10 month-old B6 mice with 6 months of AAC (ejection fraction ≤33 %). As controls, heart tissue from age-matched, sham-operated apoE-deficient and non-transgenic B6 mice were used. Hearts from 5 month-old B6 mice with 1 month of AAC were also included in the study displaying cardiac hypertrophy without failure (ejection fraction ≥50 %).

| Sample_hyb_protocol | For hybridization, 15 µg of fragmented cRNA were incubated with the chip (Mouse Genome MG430 2.0 Array) in 200 µl of the hybrization solution in a Hybridization Oven 640 (Affymetrix) for 16 h at 45 °C. GeneChips were then washed and stained using the Affymetrix Fluidics Station 450 according to the GeneChip Expression Analysis Technical Manual (Rev. 5).
